Removing the bicycle wheel and extracting the tube
You can apply this fix while keeping the wheel on the bicyckle frame, especially if it is the rear wheel, so you don't have to remove the chain from the derailleur. For the front wheel, taking it out of the bicycle is so simple as shown here, since all you have to do is slacken the nuts that hold it and pull it out of the bicycle fork.
Use a flat screwdriver to pull out the tire of the bicycle rim near the tube valve, then push the valve inside the tire to free it from the rim and take out the tube from the rim as shown. You do not need to remove the tire completely out of the rim, just extract the tube.

Fixing the punctured bicycle tube
Inflate the tube and check with your bare hand for the air leak. You will easily identify the leak by feeling the air flow on your skin.

You should use a bicycle tire repair kit (patches) like the ones shown below, but in case of urgency and if you have a car tire repair kit in hand, then you can also use it.
Spread a drop of rubber cement around the puncture, wait for a minute then apply the patch.
Make sure the area on which you spread the cement is larger than the patch.
If you're using a car tubeless tire repair kit, cut a small piece of rubber and apply it to the hole.

Build the repair patch by putting a piece of any plastic bag on top of it then squeeze it in the jaws of locking pliers.

Refitting the bicycle tube in the tire
Have your hand passing all aroung the inner side of the tire to make sure no sharp object is left there. Make sure to meticulously inspect the tire from inside since the cause of the puncture could still be there.
Start by refitting the valve then the tube all around the perimeter of the rim and in a manner to make sure it fits inside the tire.

Push the tire inside the rim, inflate it, refit the wheel to the bicycle fork and enjoy your ride!
